چکیده

Background Hepatitis B virus (HBV) infection has many faces. Precore and core promoter mutants resemble inactive carrier status. The identification of hepatitis B core antigen (HBcAg) in hepatocytes may have variable clinical significance. The present study was undertaken to detect HBcAg in chronic hepatitis B (CHB) patients and to assess the efficacy of detection system by indirect immunofluorescence (IIF) and indirect immunoperoxidase (IIP). Materials and methods The study was done in 70 chronic HBV-infected patients. Out of 70 patients, eight (11.4%) were hepatitis B e antigen (HBeAg) positive and 62 (88.57%) were HBeAg negative. Hepatitis B core antigen was detected by indirect immunofluorescence (IIF) and indirect immunoperoxidase (IIP) methods in liver tissue. Results All HBeAg positive patients expressed HBcAg by both IIF and IIP methods. Out of 62 patients with HBeAg-negative CHB, HBcAg was detected by IIF in 55 (88.7%) patients and by IIP in 51 (82.26%) patients. A positive relation among viral load and HBcAg detection was also found. This was more evident in the case of HBeAg negative patients and showed a positive relation with HBV DNA levels. Conclusion Hepatitis B core antigen can be detected using the IIF from formalin fixed paraffin block preparation and also by IIP method. This seems to reflect the magnitudes of HBV replication in CHB. How to cite this article Raihan R, Tabassum S, Al-Mahtab M, Nessa A, Jahan M, Kabir CMS, Kamal M, Aguilar JC. Expression of Hepatitis B Virus Core Antigen in Native and Fusion Forms in E. coli

DNA coding for the core antigen from hepatitis B Virus (HBcAg) was amplified, cloned and propagated in E. coli. The core protein was expressed in E. coli and the product was readily detected by Western blot. This protein can be used as a diagnostic material in serum screening tests. To increase the level of expression of this antigen in bacteria, two plasmids were constructed in which the gene ...

Significance of Hepatitis B Core Antibody as the Only Marker of Hepatitis B Virus Infection in High Risk Patients

Background and Objective: Presence of hepatitis B core antibody (anti-HBc) in the absence of hepatitis B surface antigen (HBsAg) and hepatitis B surface antibody (anti-HBs) is defined, as isolated anti-HBc. little is known about the clinical significance of the isolated anti-HBc in hepatitis B virus (HBV) infections. The aim of this study was to assess the significance of anti-HBc as the onl...

Role of HLA-B7, B8, B27, and B51 in Protection against Hepatitis B Virus Infection

Background: It has been arguedthat unprecedented degree of human leukocyte antigen (HLA) loci polymorphism within a population is requiredto avoid the devastating effects of infectious diseases. The present study was conducted to determine the associations between some of HLA classI genes and the outcome of hepatitis B virus (HBV) infection. Methods: Using sequential sampling method, 64 individ...

Priming Hepatitis B Surface (HBsAg)- and Core Antigen (HBcAg)-Specific Immune Responses by Chimeric, HBcAg with a HBsAg ‘a’ Determinant

We developed an immunogen to stimulate multivalent immunity against hepatitis B surface antigen (HBsAg) and hepatitis B core antigens (HBcAg). Immune responses specific for both HBsAg and HBcAg play an important role in controlling the infection. HBsAg-specific antibodies mediate elimination of virions at an early stage of infection and prevent the spread of virus. The immunogen was constructed...
